چکیده

We previously reported that natural killer cell stimulatory factor (NKSF), a heterodimeric lymphokine purified from the conditioned medium of human B lymphoblastoid cell lines, induces interferon y (IFN-y) production from resting peripheral blood lymphocytes (PBL) and synergizes with interleukin 2 in this activity. In this study, we show that human NKSF induces IFN-y production from both resting and activated human PBL and from freshly isolated murine splenocytes . Human T and NK cells produce IFN-,y in response to NKSF, but resting PBL require the presence of nonadherent human histocompatibility leukocyte antigens DR' (HLADR+) accessory cells to respond to NKSF. The mechanism(s) by which NKSF induces IFN-y production results in accumulation of IFN-y mRNA, is insensitive to cyclosporin A, and synergizes with those mediated by phytohemmagglutinin, phorbol diesters, anti-CD3 antibodies, and allogeneic antigens, but not by Ca2+ ionophores. The ability of NKSF to directly induce IFN-,y production and to synergize with other physiological IFN,y inducers, joined with the previously described ability to enhance lymphocyte cytotoxicity and proliferation, indicates that this lymphokine is a powerful immunopotentiating agent .

Interleukin 10 (IL-10) inhibits human lymphocyte interferon gamma- production by suppressing natural killer cell stimulatory factor/IL-12 synthesis in accessory cells

Natural killer cell stimulatory factor or interleukin 12 (NKSF/IL-12) is a heterodimeric cytokine produced by monocytes/macrophages, B cells, and possibly other accessory cell types primarily in response to bacteria or bacterial products. NKSF/IL-12 mediates pleiomorphic biological activity on T and NK cells and, alone or in synergy with other inducers, is a powerful stimulator of interferon ga...

Modulation of natural killer cell activity in mice after interferon induction: depression of activity and depression of in vitro enhancement by interferon.

Splenic natural killer (NK) cell activity of BALB/c and C3H mice was assayed after administration of the interferon inducers Escherichia coli endotoxin or Newcastle disease virus (NDV). As expected, the NK cell activity rose early in response to the interferon inducers. At 1 to 3 days after an injection of endotoxin, NK activity was hyperesponsive to interferon stimulation. At 5 to 9 days after...

Killer Cell Immunoglobulin-Like Receptors Influence the Innate and Adaptive Immune Responses

Natural killer (NK) cells are a subset of lymphocytes which play a crucial role in early innate immune response against infection and tumor transformation. Furthermore, they secrete interferon-γ (IFN-γ) and tumor necrosis factor (TNF) prompting adaptive immu-nity. NK cells distinguish the unhealthy cells from the healthy ones through an array of cell-surface receptors. Human NK cells use inhibi...

Induction of cytolytic activity and interferon-gamma production in murine natural killer cells by polymyxins B and E.

Natural killer (NK) cells are the primary effector cells of the innate immune system and have well-established roles in tumor rejection and resistance to viruses, bacteria and certain parasites. There is a need for more specific immune modulators of NK cell activity that lack the wide-ranging side effects of NK cell-stimulatory interleukins. The polycationic antibiotic polymyxin B (PMB) has bee...
